How to lock/Disable the database

I want to lock/disable the sql server 2008 database for particular time.Users should not be able to use that particular database.Is It Possible to do?

Sure.

Go to SQL Server Management Studio, right click on the database in question, on context menu go to "Tasks" and then select "Take Offline".

You probably have to force users to log off and the current active sessions will be disconnected.

Off line databases can be brought back by "Bring Online" menu option.
